What is the estimated cost (engineering hours) to modify topitcs/filtered topics/instances for a mature large-scale mission-critical project? By "to modify", I meant "to add/delect/change". What are technical factors affecting such estimation? number of P/S participants? related QoS? Thank you very much!

It sounds like you're on the right track. Are you using features like XML application creation? That might make it easier if you plan on changing/modifying your system frequently.
Another option would be to utilize routing service to modify the topics/qos/datatypes, without making any modifications to your base system.
